چکیده

blockbuster COX-2 inhibitor Vioxx (rofecoxib) as a result of an increased risk of heart attack in Vioxx users underscores the prominent role that cardiac toxicity continues to play in ensuring drug safety within the pharmaceutical industry. While the precise mechanism of Vioxx-induced cardiotoxicity is unclear, cardiac liabilities have surfaced recently for a surprising number of approved drugs that have been subsequently withdrawn.

O-3: Drug Repositioning by Merging Gene Expression Data Analysis and Cheminformatics Target Prediction Approaches

The transcriptional responses of drug treatments combined with a protein target prediction algorithm was utilised to associate compounds to biological genomic space. This enabled us to predict efficacy of compounds in cMap and LINCS against 181 databases of diseases extracted from GEO. 18/30 of top drugs predicted for leukemia (e.g. Leflunomide and Etoposide) and breast cancer (e.g. Tamoxifen a...

An advection-diffusion multi-layer porous model for stent drug delivery in coronary arteries

Arterial drug concentration distribution determines local toxicity. The safety issues dealt with Drug-Eluting Stents (DESs) reveal the needs for investigation about the effective factors contributing to fluctuations in arterial drug uptake. The current study focused on the importance of hypertension as an important and controversial risk factor among researchers on the efficacy of Heparin-Eluti...

In silico prediction of drug safety: despite progress there is abundant room for improvement.

Predictive models for drug safety are crucial for helping to avoid costly late-stage failures. We review recent work on models for genotoxicity, liver toxicity, CYP450 inhibition and cardiotoxicity. These models have improved somewhat in recent years, and research has expanded into new frontiers, such as the prediction of liver toxicity. However, much more needs to be done.:

A new classifier-based strategy for in-silico ion-channel cardiac drug safety assessment

There is currently a strong interest in using high-throughput in-vitro ion-channel screening data to make predictions regarding the cardiac toxicity potential of a new compound in both animal and human studies. A recent FDA think tank encourages the use of biophysical mathematical models of cardiac myocytes for this prediction task. However, it remains unclear whether this approach is the most ...
